Maintaining a balance between activation and suppression of immune response is critical for the immune system. Systemic Inflammatory Response Syndrome (SIRS) is an exaggerated response of the immune system and is a potentially life-threatening medical condition that is characterized by a self-harming immune response. SIRS can be initiated by an infection (then called sepsis), or by sterile causes of cell damage, such as trauma. A hallmark of both sepsis and SIRS is the massive recruitment neutrophils from the bone marrow into the circulation. Together with academic collaborators, ApoGlyx AB has demonstrated that genetic ablation of AQP9 is an effective way for interfering with SIRS, by preventing tissue infiltration of neutrophils. Based on these promising results, the aim is to decipher the structural-functional activity of AQP9 and develop a series of compounds blocking its activity. The advancement of the highly multidisciplinary field of lipid nanoparticle (LNP) research hinges largely on detailed scientific discussions. For this reason, I highly appreciate article formats that facilitate scientific discussions. In a recent Matters Arising article, I discuss some technical aspects related to a very interesting study on the biomolecular corona on LNPs. Notably, the study revealed that rats’ individual health state (obese vs. lean) significantly impacts the composition of the biomolecular corona, and consequently, the transfection potency of LNPs. Perhaps we should consider using disease-specific plasma/serum sources for in vitro LNP studies, if possible, and when relevant - especially in cases where significant alterations in the lipoprotein profile are expected. The comprehensive study presents many interesting findings, but as with many multidisciplinary-based research articles, it may be associated with some unintended limitations. In my Matters Arising article, I highlight potential technical challenges associated with this study. The beauty of this article format lies in its allowance for authors of the original article to respond to the concerns raised, fostering a rich and detailed scientific discussion. #hiring Research Associate: UMass Chan Medical School The post-doctoral scholar will lead a project focusing on understanding the complex interactions between the effector proteins of the Shigella bacteria and the host cell proteins related to the phagocytosis vacuole. This study will use in vitro models of epithelial cell cultures infected with bacteria, bacterial extracts, or purified compounds. Using chemical bridging methods to stabilize the formed complexes, the scholar will employ mass spectrometry to analyze and identify the components involved. Additionally, the post-doc will explore the impacts of these interactions on the intracellular replication of bacteria using fluorescence microscopy. This in-depth examination will offer valuable insights into the mechanisms governing the survival and growth of Shigella bacteria within host cells. By illuminating the molecular interactions at play, this research aims to enhance understanding of bacterial pathogenesis and potentially open avenues for new therapeutic approaches to combat bacterial infections.
