Our Biophysics Team at Eurofins Cerep has received an early end-of-the-year gift, NanoTemper Technologies’ cutting-edge Dianthus uHTS! Vanessa Porkolab and our Biophysics experts have already performed several pilot projects on the Dianthus uHTS, developing an in-depth understanding of the advantages of this revolutionary 1536-well compatible instrument, and how best it can be used to support client projects. With its unprecedented throughput, the High-throughput Spectral Shift (HT-SpS) offers a breakthrough in early drug discovery, making site-agnostic large screening for binders achievable! In 2023, we were one of the first CROs to offer Spectral Shift measurement using the Dianthus and we are proud to stay at the forefront of new technologies in drug discovery. Stay tuned in early 2025 for more details and our initial data coming from our HT-SpS pilot studies! Want to be the first to know more? Evolution of Leadership 2016..2024: growing closer together by diversifying roles From 2008 to 2024 we've lead NanoTemper as Co-CEOs. now after 16 years we've decided to separate our roles. Why? Because of our learnings. Especially from the last 10 years: 2014 to 2019: the time of separation. Stefan had the sole responsibility of the commercial part. I had the sole responsibility about technology & production. It worked well. Result was a company separated in two silos. 2019 to 2024: the time of re-union. Stefan and me formed the "Founders Team". We've shared responsibilites, did everything together. We've formed an Executive Team and a team of people leaders. It worked very well. The most productive time until now. Problem: doing all decision together needs a lot of coordination. A lot of overhead. It slowed us down. Werner described our problem best: "When you talk with 2 different people about the same thing, you'll understand 2 different things." Being 2 Co-CEOs wasn't - isn't - the right thing anymore. We realized, that having one single CEO, Stefan, enables us now to execute our strategy faster and better. We diversified our roles. We are now CEO & Chairman - and we are closer together than ever. Beyond bold marketing budgets - the power of 14 years personal networking: 2010, the USA: terra incognita. Huge market. How to start from zero? Is it about having a huge marketing budget? It‘s all about networking: We‘ve sold our first instrument to Cincinnati Children’s Hospital. Why? Because our Moran did his PhD there. Prof. Yi Zheng knew him. Trusted him. And so we could sell our product. When you are new in a market, you have to build trust. Especially if you sell premium products. Trust to your product. Trust to your brand. Trust to your people. Trust to you. This is a Marathon. It takes a lot of field trips. It takes a lot of time. Many years. They say it is 5 to 7 years to establish a new market. After 14 years of personal networking, I can say: A business trip to the USA now means meeting friends. Friends like Jocelyn, Ana, Dennis, Johanna, Russell, Jeff. What‘s your experience with networking? ————————— ♻️ Repost this if you find it interesting. ➕ Follow me if you want to learn more about leadership, the Honorable Entrepreneur and founding & scaling of a profitable biotech company like NanoTemper.

🎯 Protein Production Tips – Part 15: 𝐌𝐚𝐤𝐢𝐧𝐠 𝐭𝐡𝐞 𝐈𝐧𝐯𝐢𝐬𝐢𝐛𝐥𝐞 𝐕𝐢𝐬𝐢𝐛𝐥𝐞 – QC for Protein Storage As the saying goes, “making the invisible visible”—a nod to Philipp, who nailed the essence of why QC matters. The challenges with proteins don’t stop after expression or purification. Once you’ve overcome hurdles like concentration, the final step awaits: storing your precious protein. 𝐁𝐮𝐭 𝐡𝐞𝐫𝐞’𝐬 𝐭𝐡𝐞 𝐫𝐞𝐚𝐥 𝐪𝐮𝐞𝐬𝐭𝐢𝐨𝐧: How reproducible are your results when using freshly purified protein compared to protein that’s been stored for some time? And what conditions should you choose to ensure stability? Let’s unpack the considerations and tools for ensuring storage doesn’t compromise your hard work. 💡 𝐒𝐭𝐨𝐫𝐚𝐠𝐞 𝐁𝐞𝐬𝐭 𝐏𝐫𝐚𝐜𝐭𝐢𝐜𝐞𝐬 For long-term storage, it’s common to use LN₂ flash freezing, followed by storage at -80°C. (By the way, have you looked into -70°C storage? It’s an energy-saving alternative aligned with the green lab initiatives.) But here’s the catch: it’s not enough to simply decide on storage conditions—you need to test how storage impacts your protein’s quality. ✨ QC Tips for Storage Impact 1️⃣ 𝐀𝐬𝐬𝐞𝐬𝐬𝐢𝐧𝐠 𝐅𝐫𝐞𝐞𝐳𝐢𝐧𝐠 𝐄𝐟𝐟𝐞𝐜𝐭𝐬 Use nanoDSF to measure your protein’s melting temperature (Tm) before and after freezing. Compare proteins stored at 4°C and -80°C after just a few hours. If the Tm remains unchanged, you might conclude your protein is unaffected. ✅ Pro Tip: While Tm is a great start, it doesn’t always tell the full story. Subtle aggregation or unfolding may go unnoticed. 2️⃣ Looking Beyond the Surface Back in the days before Panta by NanoTemper, I relied on the Prometheus back-reflection module, which detects aggregation—a common, often invisible issue caused by freezing or suboptimal storage. ⚠️ 𝐃𝐨𝐧’𝐭 𝐁𝐞 𝐀𝐟𝐫𝐚𝐢𝐝 𝐭𝐨 𝐐𝐂 It’s easy to assume your protein is fine if it looks clear or behaves as expected in one test. But what if you’re missing something? Some might hesitate to do extensive QC because of the fear of finding an issue. Here’s my take: ❌ Having a problem isn’t the problem. Not knowing you have a problem is. Embrace the process. Testing your protein extensively now avoids headaches and inconsistencies later. 💬 Your Turn! How do you ensure your protein’s stability during storage? Have you used tools like nanoDSF or back-reflection to spot the invisible issues? San Francisco, Boston, Munich,.. Christmas all over the world. With 2 laughing eyes and 1 crying eye. Where was the best party? Everywhere. Every team celebrated differently. Every team had the best party. Because we had it as a team. Yankee swap, white elephant - sharing is caring. Learnt a lot nice games. Seen very cool dancing moves and really ugly sweaters. My first time Christmas parties in the USA. My first time missing the Christmas party in Munich. Next year is a new year. Maybe with all parties in a row. To go global means: going global. Looking forward to our party in China. 1st day at work: What's the worst thing which can happen to you? I’ve been forgotten at my first working day as PhD student. October 2005: I've arrived early in the morning, around 8:27 am, at the university. As always: a little bit early. Been very excited about my scientific origin of life project. Couldn't wait to start. How would my new colleagues be? How would my new boss be? How would the new lab be? Well - the door to the lab was closed. Waited for 30 minutes: Noone. Got more an more nervous. Toilet - lab - toilet - lab. At 8:58 am another person passed by: Lars Sonnerberg as I've learnt later. Lars was a PhD student at another team. He realised that I was nervous and asked me:"What are you waiting for?" - "Dieter" was my response. "Dieter and his team are in the mountains, they are on a team trip." WOW - my new boss has forgotten me. Left me alone - at my very first working day. My motivation: gone. My excitement: gone. I'll remember this forever. And I've told myself: This should never, never ever happen to my people! This is one reason, why we put so much emotions and energy in having a great onboarding experience for our new people. We will not forget you. Never!
