𝐓𝐢𝐦𝐛𝐞𝐫 𝐏𝐡𝐚𝐫𝐦𝐚𝐜𝐞𝐮𝐭𝐢𝐜𝐚𝐥𝐬 𝐀𝐧𝐧𝐨𝐮𝐧𝐜𝐞𝐬 𝐀𝐒𝐂𝐄𝐍𝐃 𝐓𝐫𝐢𝐚𝐥 𝐑𝐞𝐬𝐮𝐥𝐭𝐬 𝐟𝐨𝐫 𝐓𝐌𝐁-𝟎𝟎𝟏 𝐢𝐧 𝐂𝐨𝐧𝐠𝐞𝐧𝐢𝐭𝐚𝐥 𝐈𝐜𝐡𝐭𝐡𝐲𝐨𝐬𝐢𝐬 Timber Pharmaceuticals, a LEO Pharma company Timber Pharmaceuticals, Inc., a LEO Pharma company, today announced the results from the 12-week ASCEND trial, evaluating TMB-001 for moderate to severe congenital ichthyosis. Unfortunately, the trial did not show a statistically significant difference between patients treated with TMB-001 and those treated with a vehicle.' TMB-001, a topical ointment formulation of isotretinoin, was being developed for autosomal recessive congenital ichthyosis (ARCI) and X-linked recessive ichthyosis (XLRI). LEO Pharma acquired TMB-001 from Timber Pharmaceuticals in January 2024 after Timber’s Chapter 11 filing. "There are currently no topical treatments specifically developed and approved for congenital ichthyosis in the U.S., making it a challenging disease to treat," said John Koconis, CEO of Timber Pharmaceuticals. "We are disappointed by the trial results but remain committed to helping those affected by this rare condition." Kreesten Meldgaard Madsen Kreesten Meldgaard Madsen, Chief Development Officer at LEO Pharma, added: "We remain fully committed to dermatologic research and addressing unmet needs, even as not all trials succeed." The results of the trial will not have a material impact on LEO Pharma's 2024 financial results. Being that I’m on a Biologic that targets IL-23 I find it interesting how it works compared to other biologics. It’s intriguing to read even though I don’t fully understand the scientific language. I do understand the scientific names of the medications. I was on many of the IL-23 and the IL-17 mentioned in this research article. Each medication worked well, however some had greater success. It’s encouraging to me to read about all the research being conducted on psoriasis. Perhaps a cure is near. Wishful thinking on my part. “A 2020 meta-analysis comparing IL-23 inhibitor efficacy showed that risankizumab had the highest PASI90 response rates, followed by guselkumab and ustekinumab. Tildrakizumab had a lower PASI90 response rate after 10 to 16 weeks (short-term). Daniele et al compared the 2020 meta-analysis data with the structural properties of each inhibitor epitope identified in their analyses to better understand the connection between epitope structure and clinical response for IL-23 inhibitors.” I’m almost at PASI100 with two small spots remaining. I’m currently using risankizumab rzaa.

Dr. Christopher Bunick's group found that among the distinct structural properties and molecular mechanisms of action found in IL-23 inhibitors, the epitope surface area correlates most strongly with the PASI-90 scores in April Armstrong, MD MPH et al's 2020 meta-analysis in JAMA Dermatology, with risankizumab having the largest epitope SA, followed by guselkumab, ustekinumab, and lastly, tildrakizumab. #psoriasis #biologics
