𝐌𝐞𝐜𝐡𝐚𝐧𝐢𝐬𝐦 𝐨𝐟 𝐀𝐜𝐭𝐢𝐨𝐧 🌈 Vincristine is a microtubule-disrupting medication that is a member of the vinca alkaloid class of chemotherapy medications. The main way that vincristine works is through interfering with the dynamics of microtubules, especially in the M and S phases, which are crucial for cell division and other biological functions. Vincristine also inhibits the synthesis of proteins and nucleic acids by preventing the use of glutamic acid as one of its mechanisms of action. 𝐏𝐡𝐚𝐫𝐦𝐚𝐜𝐨𝐤𝐢𝐧𝐞𝐭𝐢𝐜𝐬 🌈 Vincristine is administered via an IV route and is rapidly and completely absorbed into the systemic circulation. Vincristine is extensively metabolized in the liver, mostly via the CYP (cytochrome P450) enzyme system, which includes the enzymes CYP3A4 and CYP3A5.[11] 4-O-desacetyl-vincristine is the main metabolite; it has some anticancer activity but is noticeably less effective than the original medication. N-deformyl-vincristine, N-deformyl-4-O-desacetyl-vincristine, and 4-O-desacetyl-N-deformyl-vincristine are some other minor metabolites. Genetic variations in the CYP3A4 and CYP3A5 enzymes, as well as medication interactions involving inducers or inhibitors of these enzymes, all impact vincristine metabolism. 𝐀𝐝𝐯𝐞𝐫𝐬𝐞 𝐄𝐟𝐟𝐞𝐜𝐭𝐬 🌈 Vincristine has a number of adverse effects that might vary in intensity and impact different organ systems. The cardiovascular, central nervous system, dermatological, endocrine and metabolic, gastrointestinal, genitourinary, hepatic, renal, pulmonary, neuromuscular, otic, and ocular systems are among the prominent organ systems that vincristine may adversely impact.

High insulin levels linked to increased pancreatic #cancer risk: A new study from the University of British Columbia has uncovered a direct link between high insulin levels and pancreatic cancer, shedding light on why #obesity and #Type2diabetes increase cancer risk. In a small sample of pancreatic cancer patients (n = 34), high dose Vitamin C supplementation alongside traditional chemotherapy significantly increased survival in a Phase II study. Importantly, no additional adverse events were observed in this population relative to standard of care alone. Interesting study, and another instance of non-traditional approaches to enhance the efficacy of existing treatments! Simple, cheap, and safe. Intriguing new role for GLP-1R in T cells opens doors to novel therapies in transplantation and cancer. GLP-1R, known for regulating glucose in pancreatic β cells, is also expressed by T lymphocytes during immune responses. Today is Glioblastoma Awareness Day, a crucial moment to unite in the fight against one of the most aggressive forms of brain cancer. With survival times averaging only 15-18 months from diagnosis and a five-year survival rate under 10 percent, the urgency for innovative treatments has never been greater. At BPGbio, Inc., we are actively working on developing innovative treatments for Glioblastoma Multiforme (GBM). #Akkermansia may improve the response to chimeric antigen receptor (#CAR) T-cell therapy in patients with B-cell #lymphoma. [Mouse] [Precinical study] [Report] NOTES: - Fecal samples from patients who responded to CAR T-cell therapy had a higher prevalence of #Akkermansia_muciniphila at baseline. - Akkermansia spp. supplementation resulted in greater reductions in tumor volume at 10 days , 34 days, and 46 days after CAR T-cell administration. - Overall survival was significantly improved in mice that Received Akkermansia spp. - Mice that received Akkermansia spp. had higher infiltration of CAR-T cells in the tumor at day 24 than mice that received CAR-T cells alone (30% and 3.6%, respectively). - CAR T-cell infiltration in the bone marrow at day 7 was also higher with the combination than with CAR-T cells alone (1.3% and 0.8%, respectively). - The patients who had high prevalence of Akkermansia spp. also experienced higher #tumor and bone marrow infiltration of CAR-T cells “with a higher #cytotoxic profile and a higher release of #interferon_gamma. https://lnkd.in/eVdiEfKZ

📃Scientific paper: Distinguishing benign and malignant thyroid nodules using plasma trimethylamine N-oxide, carnitine, choline and betaine Abstract: Purpose Trimethylamine N-oxide (TMAO), a gut microbiome–derived metabolite, and its precursors (carnitine, choline, betaine) have not been fully examined in relation to thyroid cancer (TC) risk. The aim of this study was to assess the value of TMAO and its precursors in diagnosis of benign and malignant thyroid nodules. Methods In this study, high-performance liquid chromatography-tandem mass spectrometry was utilized to measure the levels of plasma TMAO and its precursors (choline, carnitine, and betaine) in 215 TC patients, 63 benign thyroid nodules (BTN) patients and 148 healthy controls (HC). The distribution of levels of TMAO and its precursors among the three groups were compared by the Kruskal–Wallis test. Receiver operating characteristic curve (ROC) analysis was performed to evaluate the sensitivity, specificity, and the predictive accuracy of single and combined biomarkers. Results In comparison to HC, TC showed higher levels of TMAO and lower levels of its precursors (carnitine, choline, and betaine) (all P < 0.001). Plasma choline ( P < 0.01) and betaine ( P < 0.05) were declined in BTN than HC. The levels of carnitine ( P < 0.001) and choline ( P < 0.05) were significantly higher in BTN than that in TC group. Plasma TMAO showed lower levels in TC with lymph node metastasis (101.5 (73.1–144.5) ng/ml) than those without lymph node metastasis (131 (84.8–201) ng/ml, P < 0.05).
