In the ever-evolving field of drug discovery, speed and accuracy are crucial. The newly developed Connectivity Enhanced Structure Activity Relationship (ceSAR) framework is a game-changer, combining transcriptional signature connectivity with molecular docking to significantly enhance the efficiency of drug discovery and repurposing efforts.
Built on the extensive LINCS library of transcriptional signatures, ceSAR integrates cheminformatics and virtual screening techniques to connect small molecules with their targets.
The framework initially ranks candidate compounds based on their chemical similarity to LINCS analogs, which exhibit transcriptomic concordance with a target gene knockdown.
This process is followed by a second phase where top candidates are rescored using docking simulations, leading to a consensus ranking that improves accuracy while drastically reducing false positives.
In benchmarking tests, ceSAR has demonstrated its capability to reduce the computational time by several orders of magnitude compared to traditional docking methods, while also cutting down on false positives.
This makes ceSAR a powerful tool for virtual screening, particularly in identifying inhibitors for complex targets such as BCL2A1, a key antiapoptotic protein involved in diseases like melanoma and preterm birth-associated inflammation.
By leveraging both transcriptomic data and biophysical compatibility, ceSAR not only accelerates the identification of promising drug candidates but also democratizes drug discovery, making advanced screening tools more accessible to a broader range of researchers.
