Excitation is provided by a pulsed #laser, while point detectors (e.g. #PMT or #SPAD) or gated cameras can be used to measure the temporal decay of emission at all points in the field of view (FOV). Fitting the data with an appropriate model then provides maps of the fluorescence lifetime and amplitude in the FOV. The webinar aims to provide a basic introduction to the principles of fluorescence #emission. Then, the most common experimental methods will be briefly presented with examples of applications. 💡 This Talk is part of a series of webinars where Laserlab-Europe members introduce their techniques available to academic and industry users in the field of #recyclablematerials. It provides simultaneous illumination and detection from two opposing directions, without the need to rotate your sample. We've spent a lot of time during my PhD looking into the issue of extracting high resolution images from a complex sample that is a liver-on-a-chip. We came up with multi-modal labelled and label-free microscopy approaches (2PEF, SHG, THG, SRS) which revealed: a) Distribution of fluorescent anti-sense oligonucleotides (ASOs) from deeeeep within a chip (>150 microns) and confirmed the positive effect of GalNAc moiety on ASO hepatocyte uptake dynamics. b) Intracellular ASO trafficking is likely dependent on hepatocyte morphology. c) Chemical components associated with hepatocyte morphology were discovered potentially making hepatocyte classification and segmentation easier in future studies. It is hard to understate the importance and pervasiveness of #Fluorescence #Microscopy in the life sciences, however measuring the copy number of labeled biomolecules remains an important challenge. A new article appearing in Nature Methods takes on the degree of labeling problem, introducing a method for determining the label to target ratio. This approach allows for molecular counting and provides insights necessary for optimization of staining conditions. Unlike optical approaches, ExM physically enlarges cells and tissues by embedding them in a swellable hydrogel matrix and expanding them through controlled chemical procedures. 𝐓𝐢𝐬𝐬𝐮𝐞 𝐂𝐥𝐞𝐚𝐫𝐢𝐧𝐠: ExM can be combined with tissue clearing techniques, enabling researchers to visualize structures in complex biological samples with enhanced clarity. This is crucial for studying intact tissues and organs. 𝐈𝐬𝐨𝐭𝐫𝐨𝐩𝐢𝐜 𝐄𝐱𝐩𝐚𝐧𝐬𝐢𝐨𝐧: ExM techniques aim to isotropically expand biological samples in three dimensions, providing a more uniform enlargement compared to some optical super-resolution methods. This helps maintain the structural integrity of the sample during imaging.
