Buffer selection and how to select mobile phase for HPLC:- 1. Polarity: Consider the polarity of your analytes and match it to the mobile phase for optimal interaction. 2. Solubility: Both your analytes and the stationary phase should be sufficiently soluble in the mobile phase. 3. Viscosity: Optimal viscosity ensures proper flow rate and peak shape. As a rule, buffers should be used for a pH within +/- 1 unit of their pKa value. Within this range, buffers resist any deliberate attempts of change in pH. The buffer's capacity is at its maximum when its pH is equal to its pKa.

Wow, quite an impressive setup. They used an LTM (Low Thermal Mass) Series II for rapid heating on an Agilent 7890 with an SCD, NCD, 5975C MSD, a Capillary Flow Technology (CFT) Deans switch, two CFT splitters, and retention time locking. As if that weren't enough, they used the CFT devices to implement backflushing. Whew! 😎 Not familiar with LTM? Check it out at http://bit.ly/4gwMMFy.
