Merry Christmas! The Garvan Genomics Platform just successfully ran our first 10x Genomics on-chip multiplexing (OCM) experiment, and we didn’t make it easy for ourselves! We first isolated single nuclei from snap-frozen 2-7 mg (!!) surgical samples using the S2 Genomics Singulator. Alejandro Rios and Samira Samarfard then loaded 5000 nuclei per sample paired with a unique (coloured) gelbead barcode batch onto the new OCM chip for capture. Final library traces looked beautiful, sequencing next week for a total turnaround time of 2 weeks! Come and give it a try, benefits of this new technology include no cell loss (from hashtagging washes or genotyping requirements), species-agnostic, available for 3' and 5' assay with all the downstream (Christmas) bells and whistles (VDJ, CITE, CRISPR). Huge thanks to Nasir Shah and Blake Cochran for being our guinea pigs, here is more info on these cool samples: "Using single-nucleus RNA sequencing, we are exploring endothelial gene expression in quiescent blood vessels to better understand if we can predict how they adapt and respond to hemodynamic changes. This research could offer new insights into vascular behaviour and help develop targeted treatments for cardiovascular diseases."
