Cyclic sulfones are important pharmacophores with a wide range of pharmacological activities owing to a range of mechanisms of action. Their ability to act as conformational constraint, H-acceptor and electron-withdrawing functionality has been used to improve affinity and potency by optimizing the interactions with the target proteins. The polarity associated with sulfones helps to reduce the overall lipophilicity, which in many cases translates into improvements in ADME (absorption, distribution, metabolism and excretion) and pharmaceutical properties. Azaspirocycles, one of the representative spirocyclic compounds, are often seen in natural products and pharmaceuticals. Because of their three-dimensionality and rigidity, embedding an azaspirocycle into molecules is beneficial in medicinal chemistry. Azaspiro[2.5]octane features a spirocyclic structure where a piperidine ring is fused with a rigid cyclopropane ring. Recent research has shown that the incorporation of azaspiro[2.5]octane fragment into molecules can enhance their performance by improving biological activities and optimizing pharmacokinetics. Recently, Professor Eric N. Jacobsen's research group at Harvard University in the United States utilized a precisely designed chiral small molecule (646 Da) hydrogen bond donor (HBD) catalyst to accelerate the SN2 dealkylation step of the enantioselective Michaelis Arbuzov reaction by reproducing the geometric pre organization principle used by enzymes, thereby constructing a series of H-phosphonates with high enantioselectivity that can be derivatized in various ways. Mechanistic and computational studies have shown that although HBD reduces the reactivity of chloride nucleophiles, it can also reorganize phosphonium cations and chloride anions into a geometric structure that is prepared to enter the SN2 transition state, thereby accelerating the dealkylation step.
