I'm not a lab manager but as a senior to recent graduates or students I find a particular moment to be a hallmark: when they are able to execute an experiment (no matter how big or small) from start to finish without asking for tips, next steps, etc. When they go from reading their notes to just skimming through if they really need to. Of course, you have to make sure they really understand what they're doing and not that they're too shy to ask for help or clarifications. But usually you can tell the difference straight away!
