One thing thatt as worked for our lab, is prioritizing tests and reagents that are one demand in order to avoid wastage of resourses and also ensuring that lab personnel are oriented and are equal to the tsk when we face budget contraints so that we all are doing the right thing and work is flowing well.

Have a established protocol that you enforce to troubleshoot every test- ensure it it optimize to minimize unnecessary use of reagent while maintaining quality. Train staff to understand said protocol, all the "whys" involved so that they will have less resistance to comply. Apply sanctions when protocols are not observed on multiple occassions; share savings report with staff briefly- help them feel like part of the team in ensuring financial stability. Maintain strict reagent inventory to ensure FIFO; monitor trends/statistics to order what is needed most and when. Batch tests requested where applicable. Communicate to clients when these test are offered to facilitate samples being collected at appropriate times. Seek out investors.

Budgeting is normal for all of us, professionally, and at home. Although I have never been head of lab, I have been in charge of changing procedure to avoid overhead problems with cost-budget analysis. Actually, to my own surprise, testing out old school techniques has led to cheaper, sometimes drastically cheaper to the point of making no sense but to switch. Sometimes though, its more about innovating on ideas already seen as concrete. For example, one lab I worked in decided to go from buying Tetraphenylporphyrin to just making it. It cut the cost down so much so, that we could actually sell it if we wanted to. But the point is not about profit, but that sometimes convenience costs more than the actual compound of interest.

When facing budget constraints or issues, the most important thing to do is budgeting and prioritize. While it is difficult to try to keep equal distribution among the projects (some are close to publish, some have outside forces pushing for results and etc), it should come with the notion that once more results will be published, more grants and funds can be accessed to. Sacrifices will be made, but it is all hopefully be temporary, but it should be noted that budget constraints dont just happen, smart management and understanding where the root problem is are critical for continuous flow of the lab.
